Mean DNA concentration (mean DNA copy number per assay ± Standard Error) yielded by each of the five eDNA extraction methods from purified soil samples collected from likely yellow crazy ant nest entrances (ant nest entrance sampling event) and run using the: (a) Agra1 assay on Gieseman Road samples, (b) Agra2 assay on Gieseman Road samples, (c) Agra1 assay on Douglas samples and (d) Agra2 assay on Douglas samples. DNA yield was log10 transformed. Note the differences in y-axis scales across panels. Methods with different letters above the bars within each panel differ significantly in post-hoc tests (at P < 0.05) using Tukey HSD. Extraction methods used were field-based (Biomeme M1 Bulk Sample Prep Kit for DNA – High Concentration [Philadelphia, Pennsylvania]), CTAB (cetyltrimethylammonium bromide, Adamkewicz and Harasewych (1996)), PPLPP (preserve, precipitate, lyse, precipitate, purify method, Edmunds and Burrows (2020)), Qiagen (Qiagen DNeasy PowerSoil kit [Germantown, Maryland]) and Mu-DNA (modular-universal DNA extraction method, Sellers et al.
